What is L-carnitine really?
L-carnitine is an endogenous compound synthesized in mammals from the essential amino acids lysine and methionine. It is stored primarily in skeletal muscle and the heart (approximately 95%), while much lower concentrations are stored in the blood plasma (1). L-carnitine has attracted interest because it plays an important role in fat metabolism. The primary function of L-carnitine is the transport of long-chain fatty acids across the inner membrane of mitochondria. Through its functions in the body, it also indirectly affects carbohydrate metabolism (2).

Ingredients: L-carnitine tartrate (78.1%), capsule shell (glazing agent – hydroxypropylmethylcellulose), anti-caking agent (magnesium salts of fatty acids).

Recommended intake/preparation method:

The mechanism of action of L-carnitine is not as simple as it is commonly believed. According to a scientific publication on prolonged L-carnitine supplementation on metabolism from 2020 (Sawicka et al.), the main function of L-carnitine is to transport long-chain fatty acids to the mitochondrial matrix for their conversion into energy in the process of beta-oxidation. In addition, L-carnitine, through the reaction with acetyl-CoA (i.e. acetyl coenzyme A) and maintaining the acetyl-CoA / CoA ratio in the cell, regulates the activity of pyruvate dehydrogenase. L-carnitine also plays an important role in the regulation of metabolic pathways related to skeletal muscle protein balance: proteolysis and protein synthesis (3) and acts as an antioxidant compound, thus it can alleviate exercise-induced muscle damage (4).

L-carnitine supplementation
In the previously mentioned scientific publication (Sawicka et al.), the authors stated that L-carnitine supplementation under certain conditions may affect physical performance. They also indicated that, as reported by available studies, long-term supplementation, especially in combination with carbohydrates, may increase the total carnitine content in skeletal muscles (3).
L-carnitine and fatigue during physical exertion
From a physiological point of view, L-carnitine serves as a substrate, among others, for the synthesis of acetylcarnitine, which is necessary to maintain a real pool of free coenzyme A (CoA). It enables the continuation of the pyruvate dehydrogenase complex – theoretically, its higher flow during strenuous exercise can be expected to reduce the accumulation of lactate in the blood. As a result, it potentially helps to preserve glycogen stores and then delay premature fatigue (1).
